Direct Stamping Method And Airborne Bacteria Measurement Method
Before Measuring
1. Place the plate on top of a flat surface.
2. Hold the upper film up to inoculate the center of the lower film with 1 mL of sterilized physiological saline solution. (For a YM plate, it is recommended that Letheen broth be used.)
3. Cover the upper film.
4. Place the dedicated plastic spreader from the top of the film.
* For an AC plate, place it concave side down
*For a CC or EC plate, place it flat side down
*For a YM plate, use the spreader for YM
5. Lightly push from above, and evenly spread the sample. Do not twist or slide the spreader.
6. Take out the spreader and wait until it gels.
Gelling time direct stamping method: AC/30 min, CC, EC, YM 1 to 2 hr
Airborne bacteria measurement: AC, CC, EC, YM 1 hr
Measurement Procedure
Direct stamping method
1. Spread the upper film of the gelled Petrifilm.
2. Stick the upper film to the surface you want to study and close the film again before culturing.
Airborne bacteria measurement
1. Grip the gelled Petrifilm using a clip or the like, and fix the upper film in place with double-sided tape.
2. Leave it as is after spreading for around 15 min before culturing.
